Roberta (Arquette) is an unfilled housewife living in New Jersey who spices up her own life by reading personal ads, particularly those shared between lovers Susan (Madonna) and Jim (Robert Joy). When one of Susan's ads proposes the pair meet at a local park, Roberta attends from afar, hoping to spot on the couple in person. Roberta, however, hits her head and gets amnesia, beginning a long affair of mistaken identity as she begins to be identified as the very Susan she tried to spy on. The attention garnered wavers between positive and negative as it comes to light that someone is out to kill Susan.

A hallmark of 80s fashion and yuppie culture, DESPERATELY SEEKING SUSAN is a quintessential piece of 80s culture, and was selected for preservation in the United States National Film Registry by the Library of Congress as being "culturally, historically, or aesthetically significant".
